Streptococcus pyogenes

Crystallization (Commentary)

EC Number Crystallization (Comment) Organism
2.4.2.2 in complex with the products, ribose 1-phosphate and uracil, at 1.8 A resolution. The biological unit is a hexamer with an alpha/beta monomeric fold. Residue His169 structurally aligns with Arg168 of the Escherichia coli uridine phosphorylase structure. A second active site residue, Lys162, is not present in previously determined uridine phosphorylase structures and interacts with O2 of uracil Streptococcus pyogenes

Protein Variants

EC Number Protein Variants Comment Organism
2.4.2.2 H169A about 10% of wild-type activity Streptococcus pyogenes
2.4.2.2 H169D about 1% of wild-type activity Streptococcus pyogenes
2.4.2.2 H169N about 4% of wild-type activity Streptococcus pyogenes
2.4.2.2 K162A complete loss of activity Streptococcus pyogenes
2.4.2.2 V220D almost complete loss of activity Streptococcus pyogenes
2.4.2.2 V220E less than 0.5% of wild-type activity Streptococcus pyogenes
