To achieve the extracellular secretion of enzyme OPH in Escherichia coli, the complete opd gene along with its native signal peptide is codon-optimized and expressed in Escherichia coli BL21(DE3)pLysS. The secretion of OPH into the extracellular medium is highly affected by culture conditions, e.g. Co2+ decreases the secretion of the enzyme, while IPTG causes an enhancement in extracellular OPH secretion Flavobacterium sp.
