The glycerol activity is increased in both total pancreases and islets isolated from Aqp7-/- mice compared with those isolated from Aqp+/+ mice (tested with cell homogenates, activity assay: 50 mM Tris-HCl, pH 7.2, 5 mM ATP, 10 mM MgCl2, 100 mM KCl, 2.5 mM dithiothreitol, 4 mM glycerol, 50 microM [14C]glycerol, for 3 h at 37°C). The absence of AQP7 is associated with a moderate increase in the glycerol content of the total pancreas.
