Crystallization (Commentary)

Crystallization (Comment) Organism
purified enzyme in apoform and in complex with dTDP-3-amino-3,6-dideoxy-D-galactose, dTDP-3-acetamido-3,6-dideoxy-D-galactose, and dTDP-3-amino-3,6-dideoxy-D-glucose, hanging drop vapor diffusion method, apoenzyme crystals are grown from 100 mM MOPS, pH 7.0, and 15-18% PEG 3350, at room temperature. Internal aldimine are grown from 100 mM MOPS, pH 7.0, 17-19% PEG 3350, 200 mM KCl, and 1 mM PLP, at room temperature. Crystals of dTDP-Qui3N external aldimine complex are grown from 100 mM MOPS, pH 7.0, 12-15% PEG, 10 mM dTDP-Qui3N, and 1 mM PLP, at room temperature. Crystals of the K184A variant in the presence of dTDP-Qui3N are grown from 100 mM HEPES, pH 7.5, 18-21% PEG 5000, 10 mM dTDP-Qui3N, 200 mM KCl, and 1 mM PLP, at room temperature. Crystals of the K184A variant in the presence of dTDP-Fuc3N are grown from 100 mM MOPS, pH 7.0, 13-17% PEG 5000, 10 mM dTDP-Fuc3N, and 1 mM PLP, at room temperature. physiological function Campylobacter jejuni 81116 (Penner serotype HS:6) lipoglycan contains two dideoxyhexosamine residues, and this strain can synthesize both dTDP-3-acetamido-3,6-dideoxy-D-glucose and dTDP-3-acetamido-3,6-dideoxy-D-galactose involving enzyme WlaRG, which plays a key role in the production of these unusual sugars by functioning as a pyridoxal 5'-phosphate dependent aminotransferase activity Campylobacter jejuni
