Activating Compound

Activating Compound Comment Organism Structure
12-HPETE i.e. 12-hydroperoxy-5Z,8Z,10E,14Z-eicosatetraenoic acid, a peroxide synthesized in the platelet lipoxygenase pathway, induces TACE-mediated receptor cleavage Homo sapiens
glucose oxidase oxidative damage via glucose oxidase or hydrogen peroxide activates TACE and induces shedding of its targets, glycoprotein Ibalpha and glycoprotein V, in murine and human platelets. The TACE activation is independent of platelet activation. TACE activation induced by hydrogen peroxide is dependent on p38 mitogen-activated protein kinase signalling, whereas protein kinase C, phosphoinositide 3-kinase, and caspases are not involved. Inhibition of p38 cytoplasmic targets, phospholipase A2 and heat shock protein 27, does not prevent shedding, whereas blocking 12-lipoxygenase or Src kinase slightly inhibits TACE activation Homo sapiens
