medicine the relationship between ADAM17 and TNF polymorphisms, circulating levels of shed ADAM17 substrates, and cardiovascular risk in a prospective cohort of coronary artery disease patients is investigated. Five tag single-nucleotide polymorphisms (SNPs) of the ADAM17 gene as well as four previously described TNF SNPs are genotyped in the Atherogene Study composed of 1400 coronary artery disease patients among which 136 died from a cardiovascular cause. Soluble TNF, soluble TNF receptor 1, and soluble TNF receptor 2 concentrations are all significantly elevated in patients with future cardiovascular death, independently of other clinical/biological variables. The ADAM17-154A allele is found associated with a 14% increase of soluble TNF levels as compared to the -154C allele.
